目的体外培养胃癌细胞株SGC-7901和MGC-803,并比较两种细胞周期分布情况及侵袭能力。方法采用流式细胞术观察两株细胞的周期分布情况;采用涂有Matrigel胶的Transwell小室对胃癌细胞株SGC-7901和MGC-803进行体外侵袭实验,通过检测穿过小室的相对细胞数量比较两株细胞的侵袭能力。结果SGC-7901在G0/G1期、S期、G2/M期的分布百分比分别为(63.74±3.40)%、(30.23±2.70)%、(6.03±0.70)%;MGC-803在G0/G1期、S期、G2/M期的分布百分比分别为(62.94±6.13)%、(28.67±7.76)%、(8.39±1.69)%,两胃癌细胞株对应的周期时相分布百分比差异无统计学意义(P>0.05);检测SGC-7901和MGC-803穿过涂抹有Matrigel胶的聚碳酸酯膜的细胞数分别为65.67±6.03和94.5±3.68,差异具有统计学意义(P<0.05)。结论 MGC-803细胞的侵袭能力明显强于SGC-7901,而两株细胞在对应的周期时相分布情况则无明显差异。
Objective To culture gastric cancer cell lines SGC-7901 and MGC-803 in vitro,and compare their cell cycle and invasive ability. Methods Cell cycle properties of the cultured cells were evaluated by FCM; in vitro invasion assay using the transwells coating matrigel were performed to detect the invasive ability of gastric cancer cell lines SGC-7901 and MGC-803 by detecting their relative cell amounts of transporting through chamber. Results Distribution percentages of G0 /G1 phase,S phase and G2 /M phase were ( 63. 74 ± 3. 40 ) %,( 30. 23 ± 2. 70 ) %,( 6. 03 ± 0. 70 ) % in SGC-7901cells,and they were ( 62. 94 ± 6. 13) %; ( 28. 67 ± 7. 76) %,( 8. 39 ± 1. 69) % in MGC-803 cells,there were no significant difference about the distribution percentage of the corresponding cell cycle phases between SGC-7901 and MGC-803( P > 0. 05) ; the invasion assay using the transwells coating matrigel in vitro indicated that the invasion percentage of SGC-7901 cells was ( 65. 67 ± 6. 03 ) ,which was significantly lower than ( 94. 5 ± 3. 68) of MGC-803 cells ( P < 0. 05) . Conclusions Cell invasive ability of MGC-803 is obviously higher than that of SGC-7901 cells,while there is no significant difference in the distribution percentage of the corresponding cell cycle phases between SGC-7901 and MGC-803.
