目的研究去整合素kistrin对晶状体摘除术后兔晶状体上皮细胞(LECs)中基质金属蛋白酶2(MMP-2)蛋白表达的影响。方法对40只新西兰大白兔右眼行透明晶状体摘除术(ECLE),术毕随机分为实验组及对照组,实验组囊袋内注入0.2 mL浓度为80 ng/mL的kistrin,对照组注入等量林格液,随机取10只左眼为空白组。分别于术后14 d(实验组A组、对照组B组)及3个月(实验组C组、对照组D组)取后囊膜,以肺癌组织为阳性对照组。应用Western blotting检测兔后囊膜组织及肺癌组织中的蛋白表达量。结果空白组后囊膜MMP-2无蛋白表达,A、B、C、D组均有蛋白表达。B组MMP-2的相对灰度值为0.769±0.011,A组为0.378±0.019;D组为1.015±0.020,C组为0.361±0.013,低于D组(P=0.000);D组相对灰度值较B组升高(P=0.000);A组、C组相对灰度值差异无统计学意义(P=0.217)。结论整合素kistrin可能在晶状体摘除术后MMP-2的表达中发挥重要的作用,这可能与Ⅳ型胶原的分泌减少密切相关。

Objective To investigate the effect of disintegrin kistrin on matrix metalloprotein-2 ( MMP-2) protein expression in rabbit lens epithelial cells ( LECs) after extracapsular lens extraction( ECLE) ． Methods Forty New Zealand adult rabbits underwent ECLE in the right eye were randomly divided into experimental group，which were injected with 0． 2 ml 80 ng /ml kistrin into the capsular bag of lens，and control group，which were injected with equal amount of normal saline，with 10 left eyes as the normal blank group． Ten rabbits of experimental group and 10 of control group were executed 14 days after the operation，marked as experimental group A and control group B， and the others were killed 3 months after operation，as experimental group C and control group D，with lung cancer tissue as positive contrast group． Western blotting was used to detect the expressions of MMP-2 in posterior capsule membranes and lung cancer tissue． Results MMP-2 protein express was detected in all groups except in the normal group． MMP-2 relative gray value in group A， B，C and D were ( 0． 378 ± 0． 019) ，( 0． 769 ± 0． 011) ，( 0． 361 ± 0． 013) and ( 1． 015 ± 0． 020) ， respectively， indicating the highest value in group D( with statistical significance) ． There was no significant difference in MMP-2 relative gray value between group A and group B． Conclusion Kistrin may play an important role in the expression of MMP-2 after ECLE，which may be related to the decrease of collagen IV secretion．
