目的探讨稽留流产妇女蜕膜组织中细胞焦亡因子的表达情况及细胞焦亡对稽留流产的影响。方法采用实时荧光定量聚合酶链反应(FQ-PCR)和免疫组织化学法(IHC)分别检测稽留流产妇女(稽留流产组)和正常早期妊娠、正常人工流产妇女(正常人流组)蜕膜组织中细胞焦亡因子半胱氨酸蛋白酶-1(Caspase-1)mRNA、白介素-1β(IL-1β)mRNA及其蛋白的表达情况。结果实时FQ-PCR检测结果显示,稽留流产组蜕膜组织Caspase-1 mRNA、IL-1β mRNA相对表达量明显高于正常人流组,差异均有统计学意义(均P<0.05)。IHC结果显示,Caspase-1主要表达于细胞质内,IL-1β主要表达于细胞质和胞核内;稽留流产组蜕膜组织中的Caspase-1、IL-1β表达量均高于正常人流组,差异均有统计学意义(均P<0.05)。结论细胞焦亡因子Caspase-1和IL-1β在早期稽留流产患者蜕膜组织中的表达上调,细胞焦亡与稽留流产可能存在明显的关联。
ObjectiveTo investigate the expressions of pyroptosis factors in the decidua tissue of women with missed abortion and the effect of pyroptosis on missed abortion. MethodsThe real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) and immunohistochemistry (IHC) were used to detect the expressions of mRNA and proteins of pyroptosis factors, including Caspase-1 and interleukin-1β (IL-1β), in decidua tissues of women with missed abortion (missed abortion group) and women with normal early pregnancy and normal artificial abortion (normal artificial abortion group), respectively. ResultsThe real time FQ-PCR results showed that the relative expressions of Caspase-1 mRNA and IL-1β mRNA in decidua tissues of the missed abortion group were significantly higher than those of the normal artificial abortion group, with statistically significant differences (all P<0.05). The results of IHC showed that Caspase-1 was mainly expressed in cytoplasm and IL-1β was mainly expressed in cytoplasm and nucleus; the expressions of Caspase-1 and IL-1β in the decidua tissues of the missed abortion group were significantly higher than those in the normal artificial abortion group, with statistically significant differences (all P<0.05). ConclusionThe expressions of the pyroptosis factors Caspase-1 and IL-1β in decidua tissue of patients with early missed abortion are up-regulated, and there may be an obvious correlation between pyroptosis and missed abortion.
