目的探讨外周血hsa_circ_0105035表达水平与急性缺血性脑卒中(AIS)发生的关系,以及与脑梗死体积和临床检测指标的关联。方法纳入AIS患者310例为病例组,另选择同期年龄与病例组相匹配的无脑卒中、冠心病等心脑血管疾病史的社区人群310例为对照组,采用实时荧光定量PCR(qRT-PCR)测定所有研究对象外周血hsa_circ_0105035的表达水平,以Sanger测序和线性消化酶(RNase R)实验验证其环状结构,以核质分离实验明确其亚细胞定位。通过Image J软件计算得到AIS患者的脑梗死体积。结果hsa_circ_0105035主要分布于细胞质。病例组患者的外周血hsa_circ_0105035表达水平低于对照组,差异有统计学意义(P<0.05)。AIS患者外周血hsa_circ_0105035的表达水平与脑梗死体积呈负相关(rs=-0.127, P=0.045)。hsa_circ_0105035低表达组AIS患者的血小板体积分布宽度、乳酸脱氢酶含量高于hsa_circ_0105035高表达组,差异均有统计学意义(均P<0.05)。结论外周血hsa_circ_0105035表达可能与AIS的发生相关,并且与脑梗死体积大小有关。
ObjectiveTo investigate the correlation between the expression level of hsa_circ_0105035 in peripheral blood and the occurrence of acute ischemic stroke (AIS), as well as its correlation with cerebral infarction volume and clinical detection indicators. MethodsA total of 310 AIS patients were enrolled as the case group, while another 310 age-matched community people without a history of cardiovascular and cerebrovascular diseases such as stroke or coronary heart disease during the same period was selected as the control group. The expression levels of hsa_circ_0105035 in peripheral blood of all research subjects were detected by quantitative real-time PCR (qRT-PCR), its circular structure was verified by Sanger sequencing and linear digestive enzyme (RNase-R) experiment, and its subcellular location was clarified by nucleocytoplasmic fraction experiment. The cerebral infarction volume of AIS patients was calculated by the Image J software. ResultsHsa_circ_0105035 was mainly distributed in cytoplasm. The expression level of hsa_circ_0105035 in peripheral blood of the case group was lower than that of the control group, with statistically significant difference (P<0.05). The expression level of hsa_circ_0105035 in peripheral blood of AIS patients was negatively correlated with cerebral infarction volume (rs=-0.127, P=0.045). The platelet volume distribution width and lactate dehydrogenase content of AIS patients in the hsa_circ_0105035 low expression group were larger/higher than those in the hsa_circ_0105035 high expression group, with statistically significant differences (all P<0.05). ConclusionThe expression of hsa_circ_0105035 in peripheral blood may be correlated with the occurrence of AIS and the cerebral infarction volume.
