Study on the association between HLA-DRB1/DQB1 gene polymorphism and genetic susceptibility to breast cancer of Guangxi Zhuang women

目的探讨人类白细胞抗原(HLA)-DRB1/DQB1基因多态性与广西壮族女性乳腺癌遗传易感性的关系。方法选择90例乳腺癌患者纳入乳腺癌组,另选取同时期同一地区的85例健康体检者纳入健康体检组,所有研究对象均为广西壮族女性。采用聚合酶链反应-序列特异性引物法对两组女性的HLA-DRB1/DQB1等位基因进行分析,比较两组等位基因频率(AF)。结果两组受检者均检出13种HLA-DRB1等位基因、14种HLA-DQB1等位基因,其中乳腺癌组HLA-DRB1*01的AF为6.1%(11/180),高于健康体检组的1.8%(3/170)(Pc<0.05,OR=3.345);乳腺癌组HLA-DRB1*16的AF为3.9%(7/180),低于健康体检组的9.4%(16/170)(Pc<0.05,OR=0.418);乳腺癌组HLA-DRB1*0102的AF为4.4%(8/180),高于健康体检组的0.6%(1/170)(Pc<0.05,OR=8.519);乳腺癌组HLA-DRB1*1602的AF为2.2%(4/180),低于健康体检组的7.1%(12/170)(Pc<0.05,OR=0.144);乳腺癌组HLA-DQB1*0201的AF为38.9%(70/180),高于健康体检组的14.1%(24/170)(Pc<0.05,OR=3.381)。结论HLA-DRB1*0102、HLA-DQB1*0201、HLA-DRB1*1602基因可能与广西壮族女性乳腺癌的发生相关,HLA-DRB1*0102、HLA-DQB1*0201可能是其易感基因,而HLA-DRB1*1602可能是其保护基因。
ObjectiveTo explore the association between human leucocyte antigen (HLA)-DRB1/DQB1 gene polymorphism and genetic susceptibility to breast cancer of Guangxi Zhuang women. MethodsA total of 90 patients with breast cancer were included in the breast cancer group, and 85 healthy examinees in the same period and area were included in the health examination group. All the research objects were Guangxi Zhuang women. The HLA-DRB1/DQB1 alleles of the two groups were analyzed by polymerase chain reaction-sequence specific primer method, and the allele frequency (AF) of the two groups was compared. ResultsIn both groups, 13 HLA-DRB1 alleles and 14 HLA-DQB1 alleles were detected. The AF of HLA-DRB1*01 in the breast cancer group was 6.1% (11/180), which was higher than that in the health examination group [1.8% (3/170)] (Pc<0.05, OR=3.345). The AF of HLA-DRB1*16 in the breast cancer group was 3.9% (7/180), which was lower than that in the health examination group [9.4% (16/170)] (Pc<0.05, OR=0.418). The AF of HLA-DRB1*0102 in the breast cancer group was 4.4% (8/180), which was higher than that in the health examination group [0.6% (1/170)] (Pc<0.05, OR=8.519). The AF of HLA-DRB1*1602 in the breast cancer group was 2.2% (4/180), which was lower than that in the health examination group [7.1% (12/170)] (Pc<0.05, OR=0.144). The AF of HLA-DQB1*0201 in the breast cancer group was 38.9% (70/180), which was higher than that in the health examination group [14.1% (24/170)] (Pc<0.05, OR=3.381). ConclusionHLA-DRB1*0102, HLA-DQB1*0201 and HLA-DRB1*1602 genes may be associated with the occurrence of breast cancer in Guangxi Zhuang women, and HLA-DRB1*0102 and HLA-DQB1*0201 may be its susceptibility genes, while HLA-DRB1*1602 may be its protective gene.
