目的探讨环状RNA(circRNAs)在冠状动脉旁路移植(CABG)术后心房颤动(POAF)中的调控机制。方法从基因表达综合数据库(GEO)的数据集基因样本表达集97455(GSE97455)中获得circRNA表达谱,使用GEO2R工具在线分析GSE97455数据集中差异表达的circRNAs(DECs),然后把相应的探针名转换成国际标准circRNA名称,再利用多种生物信息学方法获取circRNAs的靶miRNAs和下游mRNA。筛选最为相关的circRNAs、miRNA及下游靶基因并对筛选结果进行RT-PCR实验验证。最后,利用Cytoscape 3.7.2构建circRNA-miRNA-hub基因调控网络图并可视化分析。结果共得到55个DECs,其中21个circRNAs表达上调,34个circRNAs表达下调。经过深入的分析,最终得到了一个由6个DECs、5个miRNAs和10个hub基因组成的circRNA-miRNA-hub基因网络图。结论本研究为circRNA和circRNA-miRNA-mRNA调控网络在POAF中的作用提供了新的视角,并经RT-PCR实验初步验证。
ObjectiveTo explore the regulatory mechanism of circular RNAs (circRNAs) in postoperative arterial fibrillation (POAF) after coronary artery bypass grafting (CABG). MethodsThe circRNA expression profile was obtained from data set gene sample expression 97455 (GSE97455) of the Gene Expression Omnibus (GEO) database, and the differential expression of the circRNAs (DECs) in the data set GSE97455 were online analyzed employing the GEO2R software. Then the relevant probe names were transferred into international standard circRNA names; furthermore, target miRNAs and downstream mRNA of circRNAs were obtained using various bioinformatics methods. The most relevant circRNAs, miRNA and downstream target genes were screened, and the screening results were verified by RT-PCR trial. Finally, the figure of circRNA-miRNA-hub gene regulatory network was constructed and the analysis was visualized by Cytoscape 3.7.2. ResultsA total of 55 DECs were obtained, among which 21 circRNAs revealed up-expression and 34 circRNAs with down-expression. After deep-going analysis, the figure of circRNA-miRNA-hub gene network consisted of 6 DECs, 5 miRNAs and 10 hub genes was finally obtained. ConclusionThe research provides a new perspective to reveal the effect of circRNA and circRNA-miRNA-mRNA regulatory network in the POAF, and is preliminarily verified by RT-PCR trial.
