目的探讨不同浓度头孢他啶对大肠杆菌生物被膜的影响。方法采用刚果红染色鉴定临床分离的大肠杆菌形成生物被膜的能力,透析后腹膜透析液-腹膜透析管系统建立大肠杆菌生物被膜,分别用连续稀释涂板细菌菌落计数和银染法染色后显微镜观察细菌生物被膜在低浓度(1×MIC)和高浓度(1×MBC)头孢他啶作用前后的变化情况。结果该大肠杆菌菌株建模7 d形成成熟生物被膜。低浓度头孢他啶对成熟生物被膜作用不显著(P>0.05),而高浓度头孢他啶能显著减少成熟生物被膜内活菌数(P<0.01),且破坏生物被膜作用明显。结论高浓度头孢他啶能破坏成熟大肠杆菌生物被膜。
Objective To observe the effect of different concentrations of Ceftazidime on Escherichia coli biofilm of peritoneal dialysis catheter in vitro. Methods The capacity of biofilm formation of clinical Escherichia coli strain was tested by the method of Congo red agar plate. Escherichia coli biofilm model was established using used peritoneal dialysis solution and the catheter. After the biofilm was treated by different concentrations of Ceftazidime,the number of viable bacteria by serial dilutions was calculated and the change of biofilm was observed using sliver staining method with optical microscopy. Results The clinical Escherichia coli strains can form biofilm and form mature biofilm at the seventh day. Compared with low concentration Ceftazidime,high concentration Ceftazidime can more significantly reduce the number of viable bacteria( P < 0. 01) ,and obviously destruct mature biofilm. Conclusion High concentration Ceftazidime can destruct mature Escherichia coli biofilm.
