目的探讨基质衍生因子-1(SDF-1)对脐带血(UCB)CD34+细胞增殖及归巢相关功能影响。方法采用流式细胞仪测定、造血祖细胞培养、结晶紫染色测定、Transwell实验等方法。结果①SDF-1在体外能通过抑制UCB CD34+细胞凋亡增强细胞生存能力;②与其他造血因子协同SDF-1可在一定程度上促进粒系祖细胞增殖;③SDF-1能提高CD34+细胞的黏附能力和迁移能力。结论 SDF-1能够应用于UCB CD34+细胞体外培养,增强UCB CD34+细胞的归巢相关功能。
Objective To study effects of stromal derived factor-1 ( SDF-1) on the proliferation and homing-related function of umbilical cord blood ( UCB) CD34cells. Methods Flow cytometry,hematopoietic progenitor cell colony culture,crystal violet staining,and Transwell test were applied. Results ( 1) SDF-1 could promote survival of UCB CD34 + cells by counteracting apoptosis. ( 2) SDF-1 in some degree could promote granulocyte progenitor cell proliferation in synergy with other hematopoietic growth factors. ( 3) SDF-1 could increase the adherent ability and migration ability of CD34 + cells. Conclusion SDF-1 can increase the homing-related ability of UCB CD34 + cells,suggesting that SDF-1 maybe helpful for the in vitro culture of UCB CD34 + cell.
