目的探讨输尿管钬激光损伤后瘢痕形成相关因子(TGF-β亚型及α-SMA)的表达情况。方法采用定量反转录-聚合酶链反应(RT-PCR)技术测定正常兔输尿管以及钬激光损伤后兔输尿管TGF-β亚型(TGF-β1、TGF-β2、TGF-β3)及α-SMA的表达水平,并进行统计学分析。结果与正常兔输尿管比较,钬激光损伤兔输尿管3周后 TGF-β1、TGF-β2及α-SMA表达量明显增高(P<0.05),而两组TGF-β3表达差异无统计学意义(P>0.05)。结论钬激光输尿管热损伤导致TGF-β亚型及α-SMA表达发生变化,与输尿管瘢痕形成及狭窄发生密切相关。
ObjectiveTo investigate the expression of scar formation-related factors [transforming growth factor β(TGF-β) subtype and α smooth muscle actin (α-SMA)] after holmium laser-caused ureteral injury. MethodsQuantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression levels of TGF-βsubtypes (TGF-β1, TGF-β2, TGF-β3) and α-SMA in normal rabbit ureter and injured rabbit ureter caused by holmium laser, and the data were statistically analyzed. ResultsCompared with normal rabbit ureter, injured rabbit ureter caused by holmium laser exhibited notably elevated expression of TGF-β1, TGF-β2 and α-SMA after 3 weeks of injury(P<0.05), but no statistically significant difference was found in TGF-β3 expression between the two groups(P>0.05). ConclusionHolmium laser-caused ureteral thermal injury results in changes in the expression of TGF-β subtype andα-SMA, which is closely related to the formation of ureter scar and the occurrence of ureter stricture.
