目的探讨人体多组织器官中EV71病毒受体SCARB2和 PSGL-1的分布特点及其意义。方法应用免疫组织化学法检测中枢神经、呼吸道、肠道、肝脏、脾脏、心脏组织EV71病毒受体SCARB2和 PSGL-1的表达。结果SCARB2在中枢神经的大脑、小脑、脑干,呼吸道的喉部、气管、支气管、细支气管、肺泡细胞、巨噬细胞以及肠道组织中呈阳性分布,肝脏呈弱阳性或阴性,脾脏、心脏则为阴性,SCARB2受体在中枢神经、呼吸道和肠道的分布明显高于肝脏、脾脏、心脏(P<0.05)。PSGL-1在中枢神经的大脑、小脑、脑干,呼吸道的喉部呈阳性分布,呼吸道的气管、支气管、肠道呈少量阳性分布,而肺泡细胞阴性,肝脏、脾脏、心脏为阴性或仅极少量表达阳性。 PSGL-1受体在中枢神经系统的分布明显高于呼吸道、肠道、肝脏、脾脏、心脏(P<0.05),PSGL-1受体在呼吸道、肠道的分布明显高于肝脏、脾脏、心脏(P<0.05)。结论在中枢神经、呼吸道和肠道表达的EV71病毒受体SCARB2和/或 PSGL-1,提示上述组织器官是EV71感染的主要部位。
ObjectiveTo explore the distribution characteristics and its significance of EV71 receptors SCARB2 and PSGL-1 in multiple human tissues and organs. MethodsThe expressions of EV71 receptor SCARB2 and PSGL-1 in the tissues of central nervous system, respiratory tract, intestinal tract, liver, spleen and heart were detected by immunohistochemistry. ResultsThe positive expression of SCARB2 was observed in the cerebrum, cerebellum and brain stem of the central nervous system, throat, trachea, bronchus, bronchioles, alveolar cells and macrophages of respiratory tract and intestinal tissues, the weakly positive or negative expression in the liver, and the negative expression in the spleen and heart. The expression levels of SCARB2 in the central nervous system, respiratory tract or intestinal tract was significantly higher in that in the liver spleen or heart(P<0.05). The positive expression of PSGL-1 was observed in the cerebrum, cerebellum and brain stem of the central nervous system and the throat of respiratory tract, the weakly positive expression in the trachea and bronchus of the respiratory tract and intestinal tract, the negative expression in the alveolar cell, and the negative or extremely weakly positive expression in the liver, spleen and heart. The expression level of PSGL-1 in the central nervous system was significantly higher than that in the respiratory tract, intestine, liver, spleen or heart(all P<0.05). The expression level of PSGL-1 in the respiratory tract or intestine was significantly higher than that in the liver, spleen or heart(all P<0.05). ConclusionThe EV71 receptor SCARB2 and/or PSGL-1 expressed in the central nervous system, respiratory tract and intestinal tract, which suggests that those organs and tissues above are the main sites EV71 infected.
