【摘要】目的探讨LKB1和P57KIP2在神经胶质瘤中的表达,研究其与临床病理特征及生物学行为的关系。方法应用免疫组化SP法检测53例神经胶质瘤和20例正常脑组织中LKB1和P57KIP2基因蛋白的表达,结合病例随访资料分析其临床意义。结果LKB1及P57KIP2蛋白在神经胶质瘤中的表达率与正常脑组织中的表达率相比较,差异具有统计学意义(P<0.05);LKB1、P57KIP2蛋白的阳性率与神经胶质瘤的WHO分级和瘤周脑水肿具有相关性 (P<0.05),与性别和年龄不相关(P>0.05);LKB1和P57KIP2蛋白在神经胶质瘤中的表达率复发组与无复发组有明显差异(P<0.05);LKB1蛋白与P57KIP2蛋白的表达呈正相关(r=-0.396,P<0.01)。结论LKB1、P57KIP2的表达与人神经胶质瘤的发生及进展相关;LKB1、P57KIP2表达的降低为神经胶质瘤发生的早期事件并且LKB1、P57KIP2低表达患者预后较差。联合检测LKB1、P57KIP2蛋白的表达对神经胶质瘤的早期诊断、判断预后有参考价值。
【Abstract】 ObjectiveTo explore the expressions of LKB1 and P57KIP2 in neurospoagioma,and to investigate the relationship of their expressions with clinicopathological features and biological behavior. MethodsThe expressions of LKB1 and P57KIP2 proteins were detected in 53 cases of neurospoagioma tissues and 30 cases of normal brain tissues using SP immunohistochemical method, and its clinical significance was analyzed on the basis of followup data. ResultsThere were significant differences in the expression rates of LKB1 and P57KIP2 proteins between neurospoagioma and normal brain tissues(P<0.05).The positive rates of LKB1 and P57KIP2 proteins correlated to WHO grade of neurospoagioma and peritumorous encephaledema(P<0.05), but did not correlate to age and gender(P>0.05). There were significant differences in the LKB1 and P57KIP2 expression rates of neurospoagioma tissues between recurrence group and nonrecurrence group(P<0.05). LKB1 protein expression positively correlated to P57KIP2 protein expression(r=-0.396,P<0.01). ConclusionThe expressions of LKB1 and P57KIP2 protein were related to the occurrence and development of human neurospoagioma. Decreased LKB1 and P57KIP2 expressions may be an early event for the incidence of neurospoagioma, and the patients with decreased LKB1 and P57KIP2 expressions have poor prognosis. Determination of LKB1 and P57KIP2 protein is valuable in the early diagnosis and prognosis assessment of neurospoagioma.
