目的研究胸腺五肽对于宫颈癌局部免疫抑制的调节作用。方法宫颈癌HeLa细胞无血清培养上清浓缩后,对其中重要的免疫抑制因子TGFβ1和VEGF进行定量,作为宫颈癌免疫抑制微环境模型,刺激正常志愿者的淋巴细胞和树突状细胞。结果宫颈癌HeLa细胞培养上清导致正常志愿者淋巴细胞中免疫抑制性的CD4+CD25+FOXP3+调节性T细胞明显增多,树突状细胞成熟受阻,而于其中加入胸腺五肽后则使该现象逆转。结论胸腺五肽在宫颈的局部应用可通过局部免疫调节,阻碍宫颈癌的进展,值得临床研究推广。
ObjectiveTo study local immune suppression regulation function of thymopentin in cervical cancer. MethodsHeLa cells cultured in serum free supernatant was concentrated and quantified on important immunosuppressive factors TGFβ1 and VEGF as cervical cancer immunosuppressive microenvironment model to stimulate normal volunteers lymphocytes and dendritic cells. ResultsCulture supernatant of cervical cancer HeLa cell caused normal volunteers ′ immunosuppressive CD4+ CD25+ FOXP3+ regulatory T cells increased significantly, dendritic cell maturation blocked. Adding thymopentin made the phenomenon reversed. ConclusionTopical application of thymopentin in cervix impedes the progress of cervical cancer by local immune regulation, and it is worthy of clinical popularization.
