目的 探讨基质金属蛋白酶2(MMP2)及基质金属蛋白酶抑制剂(TIMP2)在结肠腺瘤、结肠癌及正常结肠黏膜组织中的表达及其意义。方法 采用免疫组化法检测MMP2及TIMP2在正常大肠黏膜、结肠腺瘤及腺瘤癌变组织中的表达情况。结果 MMP2及TIMP2在正常结肠黏膜、结肠腺瘤组织及结直肠癌组织中均有表达,其中结直肠癌组织MMP2的表达量明显高于正常组织及腺瘤组织,差异有统计学意义(P<0.05),腺瘤性息肉中高级别上皮内瘤变组的MMP2表达量高于低级别组(P<0.05)。TIMP2在腺瘤组织与正常组织中的表达无差别,在结肠腺癌组织中的表达较正常组织中降低(P<0.05)。结论 MMP2及TIMP2在结肠腺瘤癌变过程中分别呈高表达、低表达,二者表达的失衡在正常黏膜-腺瘤-癌的病理形成过程中起一定作用。
Objective To investigate the expression and significance of matrix metalloproteinases 2 (MMP2) and tissue inhibitor of metalloprotease 2(TIMP2) in colonic adenoma, colonic carcinoma and normal colonic mucosa tissues. Methods The expressions of MMP2 and TIMP2 were measured using immunohistochemical staining in normal colonic mucosa, colonic adenoma and carcinoma tissues. Results MMP2 and TIMP2 were expressed in normal colonic mucosa, colonic adenoma and colorectal carcinoma tissues. The expression of MMP2 in colorectal carcinoma tissue was significantly higher than that in normal tissue and adenoma tissue (P<0.05). For the adenomatous polyps with intraepithelial neoplasia, MMP2 expression in the high grade group was higher than that in the low grade group (P<0.05). TIMP2 expression in adenoma tissue had no difference with that in normal tissue, whereas it is lower in colonic adenocarcinoma tissue compared with normal tissue (P<0.05). Conclusion The expression levels of MMP2 and TIMP2 in the process of colonic adenoma canceration are increased and decreased respectively. And this imbalance may play a role in the progression from normal mucosa to adenoma and to cancer.
